Optimize documents a tandem trapping workflow for peptide fractionation and downstream LC analysis that combines two distinct application-specific trapping functions.

The documented sequence is:

SCX trapping/fractionation -> controlled peptide release -> peptide concentration/desalting -> downstream elution and LC analysis

This is one Optimize-documented tandem workflow. It should not be treated as a universal definition of two-dimensional LC or as a general description of every 2D LC method.

This page identifies the supported application path, the two application-specific chemistry identities involved, and the confirmed product-family route. Exact cartridge sizing, holder hardware, connections, fittings, and part number are selected later.

The Documented Tandem Workflow

The workflow uses two distinct trapping stages with different functions.

The first stage uses the application-specific SCX (Strong Cation Exchange) Trap for the documented SCX trapping and fractionation step.

After controlled peptide release from that stage, the workflow uses the application-specific Peptide Concentration & Desalting Trap for peptide concentration and desalting before downstream elution and LC analysis.

At a high level, the supported sequence remains:

SCX trapping/fractionation -> controlled peptide release -> peptide concentration/desalting -> downstream elution and LC analysis

The workflow should be interpreted as a connected application path. The presence of a related chemistry name in another product family does not independently establish support for the complete workflow.

Preserve the Application-Specific Chemistry Identities

The two chemistry names in this documented workflow are application-specific identities:

SCX (Strong Cation Exchange) Trap

SCX (Strong Cation Exchange) Trap is the application-specific identity used for the documented first stage of this workflow.

It must remain distinct from generic SCX. The availability of a generic SCX phase does not, by itself, establish that the same chemistry relationship or complete 2D application is supported.

Peptide Concentration & Desalting Trap

Peptide Concentration & Desalting Trap is the application-specific identity used for the documented peptide concentration-and-desalting stage.

It should not be treated as interchangeable with generic Peptide, Peptides ES-C18, C8, C18, or another phase based solely on related terminology or historical retention comparisons.

Confirmed Product-Family Route

Reaching the supported family route does not require the customer to first choose:

  • cartridge dimensions;
  • holder hardware;
  • fittings;
  • connections;
  • exact SKU.

Check System Viability

After the application and product-family route are established, confirm that the intended method and LC system are suitable for the supported family.

Documented system constraints such as operating pressure, flow requirements, or connection requirements may determine whether the supported route is viable for a particular installation or method.

A system constraint does not independently establish another trap family as an application-equivalent substitute.

Choose the Exact Configuration

Once the application path and supported product family have been established, move to the configuration stage.

The later configuration step may require selection of:

  • exact product or part number.

Those configuration decisions should not be used as prerequisites for determining whether the documented peptide-fractionation / tandem 2D application path applies.
